RESUMO

The increased activity of PARP enzymes is associated with a deficiency of NAD+, as well as with a loss of NADPH and ATP, and consequent deterioration of the redox state in fruits. In this study, we checked whether treatment with nicotinamide (NAM) would affect PARP-1 expression and NAD+ metabolism in strawberry fruit during storage. For this purpose, strawberry fruits were treated with 10 mM NAM and co-treated with NAM and UV-C, and then stored for 5 days at 4 °C. Research showed that nicotinamide contributes to reducing oxidative stress level by reducing PARP-1 mRNA gene expression and the protein level resulting in higher NAD+ availability, as well as improving energy metabolism and NADPH levels in fruits, regardless of whether they are exposed to UV-C. The above effects cause fruits treated with nicotinamide to be characterised by higher anti-radical activity, and a lower level of reactive oxygen species in the tissue.

Pineapples are an important agricultural economic crop in Taiwan. Considerable human resources are required to protect pineapples from excessive solar radiation, which could otherwise lead to overheating and subsequent deterioration. Note that simple covering all of the fruit with a paper bag is not a viable solution, due to the fact that it makes it impossible to determine whether the fruit is ripe. This paper proposes a system by which to automate the detection of ripe pineapples. The proposed deep learning architecture enables detection regardless of lighting conditions, achieving accuracy of more than 99.27% with error of less than 2% at distances of 300 ~ 800 mm. This proposed system using an Nvidia TX2 is capable of 15 frames per second, thereby making it possible to mount the device on machines that move at walking speed.

BACKGROUND: Sunscald is a physiological disorder that occurs in many horticultural products when exposed to excessive solar radiation and high temperatures. Traditionally, sunscald is controlled using physical barriers that reflect radiation, however this practice is not always efficient. A possible alternative would be the use of chemical barriers, such as mycosporine-like amino acids (MAAs), which protect aquatic organisms against ultraviolet (UV) radiation. Thus, this study aimed to develop a lipid-based emulsion containing MAAs for using in the preharvest of horticultural products. RESULTS: Emulsions were developed using 10% (w/v) of corn oil (CO) and soybean oil (SO), carnauba wax (CW), and beeswax (BW) as lipid bases (LBs). The emulsion containing CW and ammonium hydroxide was the most stable, resembling commercial wax. Therefore, this formulation was used as the basis for the incorporation of the commercial product Helioguard™ 365, a source of MAA, in concentrations of 0%, 1%, 2%, and 4% (v/v). The MAA incorporation resulted in little modifications in the stability of the emulsion, providing an increase in the absorbance with peaks in the UV-B ranging from 280 to 300 nm. CONCLUSION: The lipid-base emulsion containing MAAs could be used as a chemical barrier to control sunscald in horticultural products. © 2021 Society of Chemical Industry.

The biosynthesis of anthocyanins has been shown to be influenced by light quality. However, the molecular mechanisms underlying the light-mediated regulation of fruit anthocyanin biosynthesis are not well understood. In this study, we analysed the effects of supplemental red and blue light on the anthocyanin biosynthesis in non-climacteric bilberry (Vaccinium myrtillus L.). After 6 days of continuous irradiation during ripening, both red and blue light elevated concentration of anthocyanins, up to 12- and 4-folds, respectively, compared to the control. Transcriptomic analysis of ripening berries showed that both light treatments up-regulated all the major anthocyanin structural genes, the key regulatory MYB transcription factors and abscisic acid (ABA) biosynthetic genes. However, higher induction of specific genes of anthocyanin and delphinidin biosynthesis alongside ABA signal perception and metabolism were found in red light. The difference in red and blue light signalling was found in 9-cis-epoxycarotenoid dioxygenase (NCED), ABA receptor pyrabactin resistance-like (PYL) and catabolic ABA-8'hydroxylase gene expression. Red light also up-regulated expression of soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) domain transporters, which may indicate involvement of these proteins in vesicular trafficking of anthocyanins during fruit ripening. Our results suggest differential signal transduction and transport mechanisms between red and blue light in ABA-regulated anthocyanin and delphinidin biosynthesis during bilberry fruit ripening.

Eggplant is rich in anthocyanins, which are thought to be highly beneficial for human health. There is no study on weighted gene co-expression network analysis (WGCNA) of anthocyanin biosynthesis in eggplant. Here, transcriptome data of 33 eggplant pericarp samples treated with light were used for WGCNA to identify significant modules. Total 13000 DEGs and 12 modules were identified, and the most significant module was associated with the secondary metabolites pathways. In addition, the hub gene SmWRKY44 with high connectivity was selected and its function was verified. The expression of SmWRKY44 showed a significant correlation with anthocyanin accumulation in the eggplant peels, leaves, and flowers. SmWRKY44-OE Arabidopsis significantly increased the accumulation of anthocyanins. Yeast two-hybrid and BiFC assays showed that SmWRKY44 could interact with SmMYB1, and it was also found that they could jointly promote the biosynthesis of anthocyanins in eggplant leaves through transient expression analysis. Our work provides a new direction for studying the molecular mechanism of light-induced anthocyanin biosynthesis in eggplant.

In this study, black geotextile inter-row mulch, a weed control practice, was applied under a semi-arid climate to attenuate solar reflection in 2015-2017, and it concurrently increased soil temperature and fruit-zone high temperature duration and decreased low temperature duration. Inter-row mulch decreased anthocyanins concentrations in grapes in 2015-2016, and consistently inhibited flavonols accumulation in 2015-2017. Correlation analysis between microclimate parameters and flavonoids concentrations reflected the importance of solar reflection, fruit-zone high and low temperature duration, heat accumulation and soil temperature to flavonoids accumulation. Basal leaf removal, a widely applied practice to increase fruit-zone light exposure, was applied to mulch-treated grapevines to investigate if increasing incident light could mitigate the impact of inter-row mulch on flavonoids, and it had limited influence on anthocyanins whereas compensated the loss of flavonols in grapes caused by inter-row mulch. Notably, inter-row mulch wines showed less red and more yellow color than controls because of lower anthocyanins concentrations.

BACKGROUND: The economic value of many fruit and vegetables is often linked to the ability to preserve such natural products. In the case of tomatoes, the manufacture of 1 kg of powder or concentrate requires the evaporation of about 16 kg water kg-1 dry matter. In the present work, the effectiveness and economic analysis of solar drying systems for drying of tomato paste with and without geothermal water heat exchanger (GWHE) were studied. RESULTS: The GWHE integration significantly increased the recorded drying temperature up to 53.41 °C, with an improvement of 11.5 °C compared to the conventional dryer. Moreover, the drying temperature inside the drying chamber exceeded ambient temperature after sunset and at night by 30.8 °C. GWHE ensured a continuous drying process, while drying time was 22 consecutive hours, compared to the basic dryer with 18 h in 3 days. Effective moisture diffusivity was found to be 1.617 × 10-9 and 1.202 × 10-9  m2  s-1 for hybrid and basic drying systems, respectively, using the slope. Three methods were used to perform economic analysis. The cumulative present worth of savings, at 23 382.6 USD, is much higher than the capital cost of the hybrid dryer, at 467 USD. The payback period is quite acceptable, at 2.21 years as compared to dryer lifetime of 15 years. Therefore, the hybrid dryer will dry tomato paste without additional charges for 12.79 years and is recommended for a large-scale tomato paste dryer. CONCLUSION: The hybrid system with GWHE ensures a continuous drying process during night-time, in addition to reduced drying time. The economic study revealed a high feasibility for the hybrid dryer, with a short payback period. © 2021 Society of Chemical Industry.

The objective of this study was to use high-energy electron beam (HEEB) treatments to find surrogate microorganisms for enteric viruses and to use the selected surrogates as proof of concept to investigate low-energy electron beam (LEEB) treatments for enteric virus inactivation at industrial scale on frozen blueberries. Six food matrices inoculated with HAV (hepatitis A virus), MNV S99 (murine norovirus), bacteriophages MS2 and Qß, and Geobacillus stearothermophilus spores were treated with HEEB at 10 MeV using 4, 8 and 16 kGy doses. G. stearothermophilus spores showed the highest inactivation on all matrices except on raisins, with a dose-dependent effect. HAV reached the maximum measurable log10 reduction (> 3.2 log10) when treated at 16 kGy on raisins. MNV showed the highest resistance of all tested microorganisms, independent of the dose, except on frozen blueberries. On frozen blueberries, freeze-dried raspberries, sesame seeds and black peppercorns, HAV showed a mean inactivation level in between those of MS2 and G. stearothermophilus. Based on this, we selected both surrogate organisms as first approximation to estimate HAV inactivation on frozen blueberries during LEEB treatment at 250 keV using 16 kGy. Reductions of 3.1 and 1.3 log10 were measured for G. stearothermophilus spores and MS2, respectively, suggesting that a minimum reduction of 1.4 log10 can be expected for HAV under the same conditions.

Rapid postharvest softening largely limits the shelf-life of persimmon (Diospyros kaki L.) fruit. Microwave is a new environmental-friendly inside-out heat-shock approach, whose effect on the fruit softening and quality has not yet been investigated. The current study applied two kinds of microwave treatments (low-power long-time, LPLT, or high-power short-time, HPST) to persimmon fruit with comparison to the hot water (HW) treatments. The results showed both microwave treatments maintained firmness, facilitated the deastringency, and increased soluble solid contents (SSC) and sugar-acid ratio of persimmon fruit. The microwave treatments reduced the cellulose and pectin degradation, and inhibited the cellulase activity, resulting in a significantly higher firmness than HW treatment and control after 2 and 4 days of storage. Moreover, application of HPST treatment down-regulated gene expression of DkPG1, DkPE2 and DkEGase1 compared with untreated fruits. These results indicated that microwave treatment is a promising soft-delaying method for the preservation of persimmon fruit.

UV-B-driven modulation of secondary metabolism in peach fruit by enhancing the biosynthesis of specific phenolic subclasses, is attracting interest among consumers. However, current literature explored the UV-B-induced metabolic changes only in peach skin subjected to direct UV-B irradiation. Accordingly, this study aimed to understand whether UV-B radiation penetrates the fruit skin and is able to induce metabolic changes also within the inner flesh. Peaches were UV-B-irradiated either 10 or 60 min, and the flesh was sampled after 24 and 36 h. Non-targeted metabolomics revealed that UV-B has a strong impact on peach flesh metabolome, determining an initial decrease after 24 h, followed by an overall increase after 36 h, particularly for terpenoids, phenylpropanoids, phytoalexins and fatty acids in the 60 min UV-B-treated samples (+150.02, +99.14, +43.79 and +25.44 log2FC, respectively). Transmittance analysis indicated that UV-B radiation does not penetrate below the skin, suggesting a possible signalling pathway between tissues.

Tomato fruits contain much organic acids and γ-aminobutyric acid (GABA) during ripening which are beneficial to human health. The effects of 4 kJ/m2 UV-C treatment on the contents of organic acids and GABA, and the expression of related genes in tomato fruits during storage at 13 °C were investigated. The results showed that UV-C treatment significantly increased the organic acids and total soluble protein contents, whereas decreased the free amino acids and glutamate contents. Besides, UV-C treatment enhanced GAD activity while reduced GABA-T activity, which resulted in accumulation of GABA. Moreover, the genes involved in the biosynthesis of organic acids and GABA were up-regulated, including CS, PEPC1, PEPC2, mMDH, cMDH, GAD1, GAD2, and GAD3, while GABA-T1 and GABA-T3 which involved in GABA degradation were obviously decreased by UV-C treatment. These results indicated that UV-C treatment might be an effective approach to accumulate organic acids and GABA during tomato fruits ripening.

BACKGROUND: In the present work we propose the use of accelerating energies (microwaves and ultrasounds) to the maceration process of sherry vinegar with citrus fruit peels (orange and lemon). For the application of microwaves, an experimental design has been developed in order to optimize the maceration conditions. To evaluate the effect of these energies on the maceration, the volatile and polyphenolic content of the samples has been analyzed, as well as their sensory characteristics. RESULTS: Orange peel provided a higher number of volatile and polyphenolic compounds to the vinegar, while lemon's yielded a greater amount of some of them. The multivariate analysis showed that the samples macerated using microwaves were the most similar to the samples macerated in the traditional way. This aspect was corroborated by the sensory analysis, which was more noticeable when orange peel was used in the macerations. CONCLUSION: Therefore, it seems that the use of microwaves to accelerate maceration is a good alternative to the traditional method of making sherry vinegars macerated with citrus peel, since it cuts down the maceration time from 3 days to just a few minutes. © 2020 Society of Chemical Industry.

In the present study, the effects of blue LED light on the regreening of citrus fruit were investigated in an in vitro system of Valencia orange flavedos. The results showed that blue LED light irradiation induced regreening in the flavedos. After four-week culture in vitro, the flavedos exhibited obviously green color in the blue LED light treatment, while the flavedos in the control were still in orange color. During the regreening process, the blue LED light treatment induced chlorophyll accumulation, and substantially altered the carotenoid composition in the flavedos. Compared with the control, the content of 9-cis-violaxanthin was decreased, while the contents of lutein, ß-carotene, and all-trans-violaxanthin were increased by blue LED light. In addition, gene expression results showed that the up-regulation of CitLCYe and down-regulation of CitLCYb2 by blue LED light led to a shift from ß,ß-branch to ß,ε-branch of the carotenoid biosynthetic pathway.

Light-emitting diodes (LEDs) have been widely used in plant factories and agricultural facilities. Different LEDs can be designed in accordance with the light quality and intensity requirements of different plants, allowing the regulation of plant growth and development, as well as metabolic processes. Blue and red lights have significant effects on anthocyanin metabolism in strawberry fruit, but their effects on other metabolites are unknown. Here, we studied the effects of blue and red lights on the metabolism and gene expression of strawberry using metabolomics combined with transcriptomics. A total of 33 differentially expressed metabolites (DEMs) and 501 differentially expressed genes (DEGs) were isolated and identified. Among these DEMs, chlorogenic acid synthesis was upregulated by the blue light compared with the red light. Co-expression network analysis of DEMs and DEGs revealed that the expression of hydroxycinnamoyl-CoA:shikimate hydroxycinnamoyltransferase (FvHCT), the main gene in the chlorogenic acid synthetic pathway, was induced by blue light. Using multi-omics-based approach, our results suggest that different LED lights have multiple effects on strawberry fruit, with blue light able to co-upregulate chlorogenic acid synthesis and FvHCT gene expression.

3Ultraviolet-C (UV-C) irradiation is known for prolonging the shelf life of many fruit by regulating different pathways. To better understand the roles of UV-C treatment in regulating the metabolic pathways in peach fruit during cold storage, transcriptomics and proteomics approaches were applied to investigate changes in peaches treated with UV-C (1.5 kJ m-2). The results showed that most differentially expressed genes (DEGs) and differentially expressed proteins (DEPs) were largely matched to carbohydrates and secondary metabolites. Further analysis found that peaches treated with UV-C exhibited higher sucrose, citric acid, malic acid, phenols, flavonoids and anthocyanins compared with untreated peaches. Proteomics and transcriptomics together indicated that changes of sugars and acids were associated with the expressions of invertase, sucrose synthase, fructokinase, malate dehydrogenase and citrate synthase. UV-C irradiation promoted the synthesis of phenols, flavonoids and anthocyanins by up-regulating expressions of phenylalanine ammonia-lyase, 4-coumarate-CoA ligase, chalcone synthase, dihydroflavonol 4-reductase and UDP-glucose:flavonoid glucosyltransferase. In summary, this research explained the general molecular mechanism of the changes of sugars, acids and phenols in peaches in response to UV-C.

Stilbenes, an active substances closely related to resistance and quality of grapes, are rarely found in natural resources. However its cumulative amount is affected by ultraviolet radiation (UV). The purpose of this study is to screen key genes in biosynthesis of stilbenes Trans-scripusin A and explore its synthetic pathway. We tested content of stilbenes with UHPLC-QQQ-MS2, results revealed that stilbenes accumulation is positively correlated with UV-B exposure time. Then, we performed transcriptome high-throughput sequencing of grapes under treatments. Results shown that 13,906 differentially expressed genes were obtained, which were mainly enriched in three major regions (ribosome, plant-pathogen interaction and biosynthesis of flavonoid). Three genes of trans-scripusin A synthesis pathway key got by combining KEGG annotation and reference gene HsCYP1B1. Phylogenetic analysis showed that SAH genes had high homology with other hydroxylase genes, and distributed in two subgroups. Gene structure analysis showed that SAH genes contained four exons, indicating that gene has low genetic diversity. Chromosome localization revealed that SAH genes were distributed on different chromosomes, in addition, the number of gene pairs between Vitis vinifera and other species was not related to genome size of other species. The expression profiles of SAH genes in different parts of Vitis vinifera L. were analyzed using qRT-PCR analysis, results indicated that expression of SAH genes be specific to fruit part. These paper provide theoretical basis for further study of polyphenols biosynthesis pathway in grape fruits. The study provides novel insights for further understanding quality of grapes response to UV radiation.

BACKGROUND: Adding value to conventional fruit juices by including prebiotic compounds into their formulation and preserving them using non-thermal, eco-friendly and safe technologies represent interesting and strategic approaches to diversify healthy and innovative food products. In this context, the effect of ultrasound-assisted processing (for 15 and 30 min, 40 kHz, 180 W) on microbiological, physicochemical, nutritional and sensory quality of prebiotic-rich strawberry juices was investigated during storage (14 days, 5 °C). RESULTS: Compared to untreated samples, the applied preservation treatments enhanced the microbiological and nutritional quality of samples by significantly reducing native microflora counts (reductions up to 1 log CFU mL-1 at day 14) and increasing the total phenolic content (by more than 25% in comparison to controls at day 14) leading to higher antioxidant capacity of prebiotic-rich strawberry juices. Ultrasound processing and prebiotic enrichment had no negative effect on sensory attributes of enriched samples, suggesting that this non-thermal technique successfully retained the fresh-like attributes of strawberry juices during their shelf-life, contributing to the good sensory stability of juices. In addition, ultrasound treatments had no detrimental impact on physicochemical quality and ascorbic acid content of enriched samples, showing similar stability to control samples during storage. CONCLUSIONS: Based on our results, ultrasound processing appears to be a promising non-thermal technique for ensuring a stable product from both microbiological and sensory points of view with improved antioxidant capacity and unaltered physicochemical quality while offering a healthier, nutritive and valuable food alternative. © 2020 Society of Chemical Industry.

Norovirus (NoV) is the leading cause of epidemic and sporadic gastroenteritis worldwide; a high number of those cases are attributed to the consumption of contaminated food. Crop producers have used several strategies to inactivate the virus present in these products and thus stop the NoV transmission chain. Physical methods such as gamma radiation show excellent results in the inactivation of bacteria, but its effect on NoV has been little studied. This study aimed to evaluate the effect of gamma radiation for NoV inactivation, and over the surface topographic characteristics of strawberry cells, as a prototype of soft fruit. A 10% suspension of GII norovirus-positive stool samples were treated with either 200 mg/L of sodium hypochlorite (NaClO) or gamma-irradiated at doses of 5, 10, 15 and 20 kilograys (kGy). Viral inactivation was determined by measuring the integrity of viral capsid using RNase A alone or in combination with proteinase K followed by RT-qPCR. The effect over cellular surface topology characteristics of the fruit was measured by atomic force microscopy (AFM) and confocal microscopy. High doses of radiation (20 kGy) were necessary to detect a significant (p < 0.05) decrease of up to 1.26 log10 viral copy number. This dose significantly (p < 0.05) raises the root means square roughness (Rq), which affects directly the quality and texture of the product. The gamma irradiation doses tested in this study were not enough to inactivate NoV. The allowed gamma irradiation doses for fresh produce does not alter the surface topology of the fruit, but they affect the content of fluorescent compounds, responsible for the antioxidant activity of the fruit.

BACKGROUND: Bushfires are becoming an increasing issue for the wine sector due to grape and vine losses and smoke taint in wine. Smoke affects vine physiology and the smoke's volatile phenols are absorbed by plants and berries, contaminating the wine. Our hypothesis was that, for the first time, unmanned aerial vehicle (UAV)-based visible images can be used to study the physiology of smoke-affected vines and to assess compromised vines. RESULTS: Procanico vines were exposed to two smoke treatments, a week apart. Gas exchanges and leaf biochemical traits were measured in the short term (30 min after smoke exposure) and in the long term (24 h after smoke exposure). Canopy damage was assessed with conventional vegetation indices (VIs) and by an innovative index derived by UAV-based visible images, the Canopy Area Health Index (CAHI). Gas exchange showed a reduction after the first smoke exposure, but the vines recovered within 24 h. The second smoke exposure led to an irreversible reduction in functional parameters. The VIs exhibited significant differences and CAHI presented a damage gradient related to bushfire nearby. CONCLUSION: The vineyard damage assessment by UAV-based visible images may represent a tool to study the physiological activity of smoke-affected vines and to quantify the loss of destroyed or damaged vines. © 2020 Society of Chemical Industry.

RESUMO

X-ray is a non-thermal technology that has shown good efficacy in reducing pathogenic and spoilage bacteria, viruses and parasites. X-ray hygiene technology resulted in a high microbial loss in numerous food products, such as dairy products, ready-to-eat shrimp, oysters, fresh products, strawberries, shredded iceberg lettuce, and spinach leaves. Some X-ray studies on food safety have shown that X-ray is an effective technology and is also an appropriate alternative to the electron beam and gamma rays, and can be used in the food industry without side effects on human health. Besides, we reviewed the X-ray effect on the nutritional value of food. Therefore in this study, we aimed to review the available pros and cons of current studies regarding X-rays' effects on the food industry.
